IMMUNOSTEP's BeadStep Assay is an immunobaead assay that uses the same principles of sandwich ELISAs, whereby a soluble substance undergoing analysis is captured between two antibodies. Bead populations have APC fluorescence. Each BeadStep assay is conjugated with a specific antibody on the surface and serves as the capture bead for that particular analyte. After incubation, biotinylated detection antibody is added and the bind to its specific analyte bound on the capture beads, thus forming capture bead-analyte-detection antibody sandwiches.  Streptavidin-phycoerythrin (SA-PE) is subsequently added, which will bind to the biotinylated detection antibodies, providing fluorescent signal with intensities in proportion to the amount of bound analyte. For each sample, the PE signal fluorescence intensity is then quantified using a flow cytometer. The concentration of the samples can be calculated by plotting the expected concentration of the standards against the MFI generated by each standard. A 4PL or 5PL algorithm is recommended for the best curve fit. When choosing a curve fitting option, try both 4 Parameter and 5 Parameter Logistic, and select the one that results in the best curve fit (highest R2 value).


This BeadStep Assay contains one vial of Working Microsphere Solution, one vial of  Detection Antibody Solution, one vial of Detection Reagent Solution, one vial of Assay Buffer, one vial of Wash Buffer and two vials of Standard protein. All solutions of this kit set have been formulated to a 1x concentration.

The Standard component is lyophilized and should be reconstituted in 1.0 mL of Assay Diluent, the standard has a protein concentration of 2,500 pg/mL. Discard unused reconstituted standard, do not store or reuse. Store lyophilized standard and other components at 4°C. Protect microsphere and the Detection Reagent from prolonged exposure to light.
