CD53, clone HI23 is a mAb expressed on B-cells, monocytes, macroph, neutroph, single CD4+/CD8+ & immature CD4-/CD8- thymocytes, and peripheral T-cells. It is effective for direct IF staining of human tissue for FCM analysis using ?1 ?g/106 cells.
This antibody reacts with the CD53-antigen, a glycoprotein of 24 kDa belonging to the tetraspanin familiy. CD53 is expressed by all leucocytes, but is absent from erythrocytes and platelets. A number of eukaryotic cell surface antigens have been shown to be related, including mammalian leukocyte antigen CD37, mammalian lysosomal membrane protein CD63, human tumour-associated antigen CD-029, and several others. These proteins are all type II membrane proteins and contain an N-terminal transmembrane (TM) domain, which acts both as a signal sequence and a membrane anchor, and 3 additional TM regions (hence the name `TM4'). The sequences contain a number of conserved cysteine residues.
CD53 may play an important role in thymopoiesis and leukocyte signal transduction and positive selection of thymocytes.
